The project has scientific novelty, since for the first time in the Republic of Kazakhstan, on a large sample, it is planned to genotype DNA samples of dairy cows according to gene loci: L-selectin, MX1, TLR4, CXCR1, BRCA1, CD14, ATP1A1; criteria such as allele frequency will be determined and genetic variants, the level of homozygosity and heterozygosity of the population, the value of the χ2 criterion, gene balance, identification of animals with a low content of somatic cells in milk, selection of animals with a complex desired genotype, selection of SNP polymorphisms that reliably influence the incidence of subclinical mastitis as a DNA marker , a Real Time PCR method will be developed for genotyping at the MX1, CD14, ATP1A1 gene loci.
Project Goals
The goal of the project is to study SNP polymorphisms in Holstein and Jersey cows associated with resistance to mastitis and the content of somatic cells in milk, optimization of genotyping methods for the L-selectin, MX1, TLR4, CXCR1, BRCA1, CD14, ATP1A1 gene loci, developed by Real Time PCR genotyping method, selection of cows with genotype resistant to breast diseases.
Expected and/or achieved results
Based on the results of the scientific project, the project implementers will publish: - at least 2 (two) articles in journals from the first three quartiles according to the impact factor in the Web of Science database or having a CiteScore percentile in the Scopus database of at least 50. According to the stated topic, there will be 200 Holstein cows and 200 Jersey cows were genotyped for the following gene loci: L-selectin, MX1, TLR4, CXCR1, BRCA1, CD14, ATP1A1 loci, alleles of which have an associative effect on the incidence of mastitis and on the content of somatic cells in milk. In the theoretical aspect, interesting results, in our opinion, will be obtained: the prevalence of genetic variants, allele frequency, gene balance, the value of the χ2 criterion, gene balance at the studied gene loci in the studied animals. The study is of an experimental nature; as a result of genotyping, animals with the desired genotype, more susceptible to mammary gland pathologies, with a low content of somatic cells in milk will be identified, individuals with a complex desired genotype, which are more resistant to mastitis, will be identified. Based on the results of the study, genes whose alleles are reliably associated with the content of somatic cells in milk will be determined, the identified SNP polymorphisms will be recommended as DNA markers of resistance to mastitis in dairy cows, and these DNA markers will be included in breeding programs for dairy cattle in the Republic of Kazakhstan. Genotyping of cows by gene loci (L-selectin, MX1, TLR4, CXCR1, BRCA1) will be carried out using the existing PCR RFLP method; for genotyping DNA samples by the ATP1A1 gene locus, the search for the necessary restriction enzyme will be carried out using the NEBcutter program (https://nc3. neb.com/NEBcutter/prj/p0JkXBs6219-PGDSj_Unnamed ) and for genotyping cows at the MX1, CD14, ATP1A1 gene loci, a Real Time PCR method will be developed, which reduces the duration of molecular genetic analysis of samples. Based on the results obtained, two applications will be filed to obtain an innovative patent for an invention of the Republic of Kazakhstan. The project has a socio-economic effect, since providing the population of the Almaty metropolis with environmentally friendly products is a state task of the agro-industrial sector of the economy, the implementation of the research results will reduce the incidence of mastitis in dairy cattle, animals with a complex desired genotype will be selected. Previously, we sent a proposal to the Ministry of Agriculture of the Republic of Kazakhstan to create a Republican Center for genetic monitoring of breeding animals for carriage of such hidden genetic defects as fertility haplotypes HH1, HH2, HH3, HH4, HH5, HH6, HCD, BC, BLAD, CVM, DUMPS, FXID , JH1, JH5, subfertility syndrome in bulls, based on the results obtained, we will recommend individual SNP polymorphisms as DNA markers of resistance to mastitis. The results of the study will be introduced into the educational process in the elective discipline “Molecular genetic diagnostic methods in veterinary medicine” for students of the educational program “Veterinary Medicine”.
Information for potential users
An analysis of domestic literature shows that there are publications by Kazakhstani scientists on the study of the associative influence of gene alleles associated with the content of somatic cells and the incidence of mastitis. We believe that the study of SNP polymorphisms associated with the content of somatic cells in milk, the identification of cows resistant to mastitis, the development of molecular genetic methods for genotyping cows at the loci of the L-selectin, MX1, TLR4, CXCR1, BRCA1, CD14, ATP1A1 genes are an urgent problem of veterinary science , determining the number of somatic cells and the residual amount of antibiotics in milk is of great practical importance. In our country, for the first time on a large sample, it is planned to genotype DNA samples of dairy cows at seven gene loci; criteria such as the frequency of alleles and genetic variants, the level of homozygosity and heterozygosity of the population, the value of the χ2 criterion, gene balance, identification of animals with a low content of somatic cells in milk, selection of animals with a complex desired genotype, selection of SNP polymorphisms that reliably influence the incidence of subclinical mastitis as a DNA marker, a Real Time PCR method will be developed testing for the MX1, CD14, ATP1A1 gene loci.
